In this paper, based on fieldwork and herbaria surveys, new data concerning the presence of 32 native and alien vascular species for Sicily (Italy) are provided. Among the native species, the occurrence of the following taxa is reported for the first time or confirmed after many decades of non-observation: Aira multiculmis, Arum maculatum, Carex flacca subsp. flacca, Mentha longifolia, Oxybasis chenopodioides, Najas minor and Xiphion junceum. Furthermore, we document the presence of three native species (Cornus mas, Juncus foliosus and Limonium avei) that, despite being repeatedly observed in Sicily and reported in the literature, are inexplicably omitted by the most recent authoritative checklists regarding the flora of Italy. Finally, fifteen alien species new to Sicily (including one new to Europe, i.e., Pyrus betulifolia) are reported and seven poorly documented allochthonous taxa are confirmed for the island, and for two of them, a status change is proposed. These new or confirmed records allow us to better define the European and national distribution of the targeted taxa and offer new insights on the native and alien flora of Sicily.

Rapid environmental change is forcing populations into environments where plasticity will no longer maintain fitness. When populations are exposed to novel environments, evolutionary theory predicts that genetic variation in fitness will increase and should be associated with genetic differences in plasticity. If true, then genetic variation in plasticity can increase adaptive potential in novel environments, and population persistence via evolutionary rescue is more likely. To test whether genetic variation in fitness increases in novel environments and is associated with plasticity, we transplanted 8149 clones of 314 genotypes of a Sicilian daisy (Senecio chrysanthemifolius) within and outside its native range, and quantified genetic variation in fitness, and plasticity in leaf traits and gene expression. Although mean fitness declined by 87% in the novel environment, genetic variance in fitness increased threefold and was correlated with plasticity in leaf traits. High fitness genotypes showed greater plasticity in gene expression, but lower plasticity in most leaf traits. Interestingly, genotypes with the highest fitness in the novel environment had the lowest fitness at the native site. These results suggest that standing genetic variation in plasticity could help populations to persist and adapt to novel environments, despite remaining hidden in native environments.

Crocus is a taxonomically complex genus, characterized by high intra- and inter-specific variability. Five wild Crocus taxa - Crocus biflorus, C. longiflorus, C. neapolitanus, C. siculus and C. thomasii from three different areas of Southern Italy (Calabria, Basilicata and Sicily) have been investigated. We considered both flower morphological traits (inner and outer perigone segments, style branches and anthers) and the chemical profile of specialised metabolites (apocarotenoids: crocins; flavonoids: flavonols) in style branches, which were determined by high-performance liquid chromatography, coupled with a diode array detector and mass spectrometry (HPLC-UV-DAD-ESI-MS). Saffron (C. sativus) was also included as the 'control' species. The length of perigone tube, outer and inner tepals, anthers and style branches gave the highest contribution to the discrimination of different taxa. Concerning the specialised metabolite profiles, 20 flavonols and 24 crocins have been identified and quantified in the Crocus extracts and used to discriminate among samples, confirming that Crocus taxa can be considered as an important natural source of these substances. The chemical profiles of the different populations showed some distinctive qualitative and quantitative patterns that contributed to a certain degree of discrimination among species, in fact, flavonoids content range is comprised between 2.7 and 145.9 mg/g, whereas crocins range between 19.8 and 604.0 mg/g. It is thus hypothesized that the combination of morphological and phytochemical screenings may be an effective methodology to characterize wild Crocus genotypes from Southern Italy, also in comparison to C. sativus (saffron).

The evolution of plastic responses to external cues allows species to maintain fitness in response to the environmental variations they regularly experience. However, it remains unclear how plasticity evolves during adaptation. To test whether distinct patterns of plasticity are associated with adaptive divergence, we quantified plasticity for two closely related but ecologically divergent Sicilian daisy species (Senecio, Asteraceae). We sampled 40 representative genotypes of each species from their native range on Mt. Etna and then reciprocally transplanted multiple clones of each genotype into four field sites along an elevational gradient that included the native elevational range of each species, and two intermediate elevations. At each elevation, we quantified survival and measured leaf traits that included investment (specific leaf area), morphology, chlorophyll fluorescence, pigment content, and gene expression. Traits and differentially expressed genes that changed with elevation in one species often showed little changes in the other species, or changed in the opposite direction. As evidence of adaptive divergence, both species performed better at their native site and better than the species from the other habitat. Adaptive divergence is, therefore, associated with the evolution of distinct plastic responses to environmental variation, despite these two species sharing a recent common ancestor.

Investigations on seed biology and ecology are of major importance for the conservation of threatened plants, both providing baseline information and suggesting practical approaches. In our study, we focused on the germination behavior of Silene hicesiae Brullo & Signor., a narrow endemic species to Panarea and Alicudi (Aeolian Archipelago, Italy), as well as one of the 50 most threatened Mediterranean island plants. Specifically, the effects of temperature, light, seed age, seed source, and collection year were evaluated; in addition, threshold temperatures and thermal-time parameters were estimated. The thermal range for fresh seed germination resulted between 5 and 15 °C, reaching up to 20 and 25 °C at increasing seed age, with 30 °C being clearly beyond the ceiling temperature. This behavior indicates that fresh seeds exhibit the Type 1 non-deep physiological dormancy, and that germination is regulated by conditional dormancy. This dormancy syndrome emerged as a highly efficient adaptation strategy for this species and, together with thermo-inhibition, would allow seeds to counteract or take advantage of Mediterranean environmental conditions. The comparison between the wild Panarea population and the corresponding ex situ cultivated progeny has enabled the identification of the latter as a suitable seed source for sustainable in situ reinforcement actions, at least in the short-term; indeed, plant cultivation for a single generation did not produce significant modifications in the germination behavior of the offspring.

A synthetic and updated overview about the vascular flora and vegetation of the Island of Capo Passero (SE-Sicily) is provided. These data issue from two series of field surveys-the first carried out between 1997 and 2000, and the second between 2005 and 2019 and mostly focused on refining and implementing vegetation data. The current islet's flora consists of 269 taxa, of which 149 (58%) are annual plants. The Mediterranean species are largely prevailing, 108 (40%) of which have a strictly Mediterranean biogeographical status. The comparison with a species list published in 1919 and updated in 1957 suggest that, despite the overall prevalence of anemochorous taxa, the vertebrate fauna represents an important vector for the plant colonization of the island, while the immigration of myrmechocorous taxa does not compensate the extinction rate. As many as 202 phytosociological relevés, 191 of which issue from original recent field surveys, enabled identifying 12 different plant communities. The comparison with a vegetation map published in 1965 suggests a strong reduction in dune habitats (2120 and 2210 according to EU 'Habitats' Directive 92/43), as well as a deep disruption in the succession typical of the local psammophilous vegetation series. In order to preserve rare, endangered and protected plant species (such as Aeluropus lagopoides, Cichorium spinosum, Limonium hyblaeum, L. syracusanum, Poterium spinosum, Senecio pygmaeus and Spergularia heldreichii) and to stop the ongoing habitat degradation, urgent and effective conservation measures should be adopted for this tiny, yet precious islet.

Most legumes are well-known for the physical dormancy of their seeds; hence, the implementation of appropriate scarification techniques is essential for introducing new legume crops within agricultural systems. This study investigated morpho-anatomical traits and dormancy-breaking requirements in two taxa of the genus Bituminaria: the widespread B. bituminosa and the point endemic B. basaltica. As the species under investigation show monospermic indehiscent legumes, pods were used in this research. We performed pod trait measurements, light microscopy observations on the seed coat anatomical structure, and germination tests after mechanical, thermal, and chemical scarification treatments for seed dormancy breaking. Moreover, germination performance at different pod maturity stages and storage times was tested. Differences in morpho-anatomical traits were found, with B. basaltica having a thicker palisade cell layer and B. bituminosa showing larger pods. All of the scarification treatments proved to be able to break physical dormancy, with mechanical and chemical scarification being the most effective methods in both species. Nevertheless, dormancy-breaking treatments performed better in B. bituminosa. Seeds at early pod maturity stages showed higher germination capacity in both species. Overall, this research provided background knowledge on seed collection time, storage strategy, and effective pre-sowing treatment, which might contribute to enhance propagation and use of Bituminaria species for multiple purposes. Under this perspective, the future characterization of additional Bituminaria genetic resources from other Mediterranean populations will have remarkable importance.

As climate change transforms seasonal patterns of temperature and precipitation, germination success at marginal temperatures will become critical for the long-term persistence of many plant species and communities. If populations vary in their environmental sensitivity to marginal temperatures across a species' geographical range, populations that respond better to future environmental extremes are likely to be critical for maintaining ecological resilience of the species. Using seeds from two to six populations for each of nine species of Mediterranean plants, we characterized patterns of among-population variation in environmental sensitivity by quantifying genotype-by-environment interactions (G × E) for germination success at temperature extremes, and under two light regimes representing conditions below and above the soil surface. For eight of nine species tested at hot and cold marginal temperatures, we observed substantial among-population variation in environmental sensitivity for germination success, and this often depended on the light treatment. Importantly, different populations often performed best at different environmental extremes. Our results demonstrate that ongoing changes in temperature regime will affect the phenology, fitness, and demography of different populations within the same species differently. We show that quantifying patterns of G × E for multiple populations, and understanding how such patterns arise, can test mechanisms that promote ecological resilience.

This study aims to explore the effect of environmental factors (temperature, light, storage time) on germination response and dormancy patterns in eight Mediterranean native wildplants, belonging to the Euphorbia L. genus. In detail, we considered E. amygdaloides subsp. arbuscula, E. bivonae subsp. bivonae, E. ceratocarpa, E. characias, E. dendroides, E. melapetala, E. myrsinites, and E. rigida. We collected seeds from natural plant populations and performed germination assays in climatic chambers at seven constant temperatures (from 5 to 35°C, with 5°C increments), and four fluctuating temperature regimes (8/15, 8/20, 8/25, and 8/30°C, with a 12/12 hr thermoperiod). Germination assays were set up both in dark (D) and in light/dark conditions (L/D, 12/12 hr photoperiod), after short and long seed storage (SS around 30 days and LS around 150 days). For all these species, except E. amygdaloides subsp. arbuscula, results show that the final germinated proportions were improved by a long storage period (>150 days), which supports the existence of nondeep physiological dormancy. Optimal temperature levels ranged from 14.3 to 21.3°C and base temperatures ranged from 5.6 to 12.1°C, while ceiling temperatures from 25.6 to 34.7°C. For none of these species, germinations were favored by an alternating daily temperature regime, while in several instances, germinations were quicker and more complete in darkness, than in an alternating light/dark regime. In some instances, extreme temperature levels (5 and 30°C) induced dormancy and germinations did not resume when seeds were exposed at optimal temperature levels. Results are discussed in terms of the dynamics of emergences and how this might be affected by climate changes.

RESUMO

A new species of Bituminaria is described and illustrated: Bituminaria antiatlantica Brullo, C. Brullo, Cambria, Cristaudo & Giusso, sp. nov., which is endemic to Anti-Atlas Mountains (Morocco). It is a true chasmophyte, characterized by a suffruticose habit, several woody branches, leaflets coriaceous, rounded to ovate, small, few-flowered inflorescences and corolla pale coloured.

BACKGROUND AND AIMS: In the Mediterranean basin, the Italian peninsula has been suggested to be one of the most important glacial refugia for temperate tree species. The orchid genus Epipactis is widely represented in the Italian peninsula by widespread species and several endemic, localized taxa, including selfing and outcrossing taxa. Here the phylogenetic and phylogeographic relationships in a group of closely related taxa in Epipactis are investigated with the aim of understanding the role of this refugial area for cladogenesis and speciation in herbaceous species, such as terrestrial orchids. METHODS: Ribosomal DNA (rDNA) was employed to assess phylogenetic relationships, and plastid sequence variation in the rbcL-accD spacer was used to reveal phylogeographic patterns among plastid haplotypes using a parsimony network. KEY RESULTS: Low genetic variation and shared ribotypes were detected in rDNA, whereas high levels of sequence variation and a strong phylogeographic structure were found in the examined plastid region. The parsimony plastid haplotype network identified two main haplotype groups, one including E. atrorubens/microphylla/muelleri/leptochila and the other including all accessions of E. helleborine and several localized and endemic taxa, with a combination of widespread and rare haplotypes detected across the Italian peninsula. A greater genetic divergence separated the Italian and other European accessions of E. helleborine. CONCLUSIONS: Phylogenetic and phylogeographic patterns support a working hypothesis in which the Italian peninsula has only recently been colonized by Epipactis, probably during the most recent phase of the Quaternary age and, nevertheless, it acted as a remarkable centre of diversification for this orchid lineage. Changes in pollination strategy and recurrent shifts in mating system (from allogamy to autogamy) could have represented the mechanism promoting this rapid diversification and the observed high taxonomic complexity detected in the E. helleborine species complex.
